I;ve see a picture somewhere with the big gromet (rubber plug) removed to allow a pressure test gauge to be connected.... something to do with an auto box test?
Not sure about that, the connection to read the box pressure is at the rear of the box under where the rear extension bolts on, it's a small tapered plug.

According to my manual its for checking the hydraulic pressure on the "overdrive" manual box?
Yes, it is to allow for pressure check on the overdrive operating valve on an 'A' type overdrive. The J type is accessed from below. See photo p88 Haynes or section 40.20.01 in the workshop manual.
