The A4 runs, very roughly, parallel to the M4 but south of it. If you're looking for the easiest way then I'd get on the M4 for just a short distance and come off at J18 (nice pub nearby) and head south for just a couple of miles until you can turn left onto the A420. Follow this to Chippenham and from there get on the A4 to Reading - there'll even be a good photo opportunity for you just east of Calne where there's one of those big chalk white horses !
An alternative start, to avoid the M4 completely, is the A4174 - a sort of half ring road around Bristol - that starts in Filton and goes round to join up with the A420.
