Hi Dave,
I did mine when I fitted new UJ's to my driveshafts. It is easy to fit grease nipples to the female half of the driveshafts while they are on the bench, and then just top up with constant velocity joint grease as required. You will see the rubber gaiter start to expand slightly as the grease fills the splines and spills out into the gaiter.
The only possible problem is the danger of blowing off the blanking end cap which sits in the end of the female half of the driveshaft, when you fill it with grease. It will be obvious where it i when the drivesahft is on the bench - just make sure you check the end cap is held firmly in its position before you start pumping grease into the shaft.
